Education in the field of molecular genetics in the Genetic laboratory at Crop Science Department at Agricultural Institute of Slovenia in Ljubljana within the bilateral project

Sonja Rašeta, MSc, senior researcher, was on education in the field of molecular genetics in the Genetic laboratory at Crop Science Department at Agricultural Institute of Slovenia in Ljubljana from 21st of February to 4th of March, 2022 through the bilateral project "Characterization of leafy kale (Brassica oleracea var. acephala L.) for its conservation and sustainable use" between Bosnia and Herzegovina and Slovenia.

Protocols and methodology used for the molecular characterization of leafy kale were already established in the Genetic laboratory at Crop Science Department at Agricultural Institute of Slovenia in Ljubljana according to Pipan and Meglič, 2019; Meglič and Pipan, 2018; Pipan et al., 2013, as well as other inhouse protocols.
 

DNA isolation was done from 48 samples of young leaves of leafy kale using Qiagen Retsch Tissue Lyser and Applied Biosystems MagMAX Express and Qiagen Biosprint DNA Plant Kit.
 
DNA quantification was done usinf Life Technologies Qubit 3.0 fluorometer and Qubit dsDNA Broad Range Assay Kit.
DNA visualization on agarose electrophoresis was done using Syngene GeneSnap software.
PCR amplification was done using Applied Biosystems by Thermo Fisher Scientific - Verity and visualization of the PCR product was done on agarose electrophoresis.

The next step - fragment analysis is to be performed by a research team from Slovenia.
