Evaluation and genetic caracterization grape gene pool in Herzegovina

 Grapevine gene pool in Herzegovina is poorly studied regarding genetic analysis. There are a number of old grapevine cultivars having different names that could have same genotype; or cultivars with different genotypes could have same name. At the same time origin of these cultivars stay unknown and in most cases parents of the cultivars are not known. In order to identify existing vineyards and nurseries of grapevine as well as individual producers having old cultivars in their plantations there have been done terrain survey. There have been identified 18 vines of old cultivars inventoried at 5 different locations. Materials for molecular analysis were taken as well as grapes for ampelographic analysis (for Dalmatinka, Plavka, Radovača, Rezaklija, Surac, Surac plavi and Žilavka) and filometric characteristics of leaves (Blatina i Žilavka). Genetic characterization was done on all collected samples by amplification at 22 microsatellite loci [VVS2 (Thomas and Scott, 1993), VVMD5 and VVMD7 (Bowers et al., 1996), VVMD25, VVMD27, VVMD28 and VVMD32 (Bowers et al., 1999), VrZAG62 and VrZAG79 (Sefc et al., 1999), VChr3a, VChr5a, VChr5b, VChr5c, VChr7b, VChr8a, VChr8b, VChr9a, VChr13a, VChr15a, VChr18a, VChr19a and VChr19b (Cipriani et al., 2010, Cipriani et al., 2008)]. By analysis it was determined one group of cultivars having same genetic profiles at all analyzed loci Kadarun (Maksimović) = Kadarun (Runjevac) = Surac (Runjevac). Samples having same names always had same genetic profiles at all analyzed loci [Kadarun (Maksimović) = Kadarun (Runjevac), Plavka (Runjevac) = Plavka (Vujović); Radovača (Maksimović) = Radovača (Bakoč) i Žilavka (Maksimović) = Žilavka (Bakoč) = Žilavka (Runjevac)]. When comparing genetic profiles of analyzed samples with the genetic profiles of already genotyped international cultivars there have been established additional groups of cultivars having same genetic profiles: Dalmatinka = Alicante Bouchet while sample NPS VI had same genetic profile as a modern hybrid Diretta Bianca. There have been established 13 unique genotypes within the 18 analyzed samples of grapevines: Dalmatinka (=Alicante Bouchet), Drenak, Kadarun (Maksimović) = Kadarun (Runjevac) = Surac (Runjevac), NN VI, NPS VI (=Diretta Bianca), Plavka (Runjevac) = Plavka (Vujović), Radovača (Maksimović) = Radovača (Bakoč), Rezaklija, Smederevka, Surac, Surac Plavi and Žilavka (Maksimović) = Žilavka (Bakoč) = Žilavka (Runjevac). Ampelographic evaluation and genetic analysis represent important results that can contribute establishment of the collection vineyard of old grapevine cultivars of Republika Srpska.
